二、 许雪梅课题组

1. 课题组简介

疫苗是因传染性疾病的预防所需而问世的,包括传统的急性传染病以及慢性传染病(如HBVHPV等感染相关疾病),在对抗传染病的中起着十分重要的作用。目前上市疫苗还存在一些缺陷,主要是保护范围不够理想,如HPV黏膜致癌型约20多型、黏膜低危型约12型,但目前上市的4HPV病毒样颗粒(VLP)蛋白多价疫苗中,最高的9价苗仅能保护7种致癌型、2种低危型;肠道病毒EV71手足口病灭活苗17年在我国问世应用后,手足口病的发病率并没有下降,目前尚无多价手足口病疫苗的问世;四价流感病毒的保护效率也难以满足流感预防的要求,且会出现预测病毒株与现实流行株不一致,导致疫苗脱靶。本课题组的主要研究方向,首先是采用基因工程重组蛋白技术,研制通用型HPV嵌合病毒样颗粒(cVLP)疫苗、多价手足口病病毒样颗粒疫苗及通用型流感病毒疫苗。目前HPV通用疫苗的专利技术已经转让,该技术为表面展示保守表位的HPV cVLP疫苗,动物实验结果表明多种cVLP联合应用具有协同加强作用,中和不仅没有交叉抑制现象,同时还可协同强化针对部分交叉和型别的中和抗体滴度,具有广谱的交叉中和活性,可至少中和24余种HPV型别,包括所有12种高危型、大部分少见的致癌型,同时还2种主要低危型和4种皮肤型;手足口病多价疫苗及流感病毒通用型疫苗正在进行中。其次是开展各种疫苗佐剂的研究,包括传统的铝佐剂、具有TLR刺激活性的新型小分子化合物佐剂、乳剂及新型复合佐剂等的研究。HPV治疗性疫苗,主要选择与病毒感染及感染相关恶性表型维持必需的癌蛋白为靶抗原,定点突变后消除其转化活性,进行融合各种免疫刺激分子的基因疫苗研究。

三、 王晨轩课题组

1. 课题组简介

研究方向:蛋白质的正确折叠与组装是维持人体生理机能的必需。肽链通过精密地调控分子间相互作用自组装形成有序三维分子结构,进一步组装、聚集、相分离,形成执行生理功能的分子机器与无膜细胞器。蛋白质组装稳态失衡是一系列退行性疾病共同的病理生理基础。研究蛋白质组装过程与结构,认识自组装过程中的分子间相互作用的本质与规律,实现对自组装过程与功能的有效调控对蛋白质组装相关疾病的预防和治疗有重要意义,从经典药物治疗到相关新型药物治疗的兴起,从微观层面对发病分子机理的阐述到宏观层面治疗模式的创新,新的认识将在未来改变相关疾病的诊治模式,为患者改善预后带来新的机遇。王晨轩课题组遵循“认识组装-调控组装-利用组装”的研究思路,发展扫描隧道显微术、原子力探针显微术、核磁共振波谱技术、X-射线单晶衍射、光散射等新颖的物理、物理化学方法用于生物大分子的结构测定,在单分子水平理解与重大疾病发生、发展密切相关的蛋白、多肽的折叠、组装结构和过程,认识蛋白、多肽折叠组装过程中的相互作用本质与调控规律。

王晨轩,1986年出生,博士,研究员,士生导师,中国医学科学院基础医学研究所课题组组长。2004-2008年,兰州大学国家理科化学基地班,本科。2008-2013年,中国科学院国家纳米科学中心与清华大学化学系联合培养研究生,物理化学专业。导师王琛研究员、杨延莲研究员、李艳梅教授。研究方向:淀粉样蛋白折叠结构的研究及多肽-药物相互作用。2013年获清华大学理学博士学位。2010年,丹麦奥胡斯大学多学科交叉纳米中心-物理与天文系访问学者。合作导师丹麦皇家科学院院士、中国科学院外籍院士Flemming Besenbacher,董明东教授。研究方向:膜蛋白的超高分辨原子力探针显微术的单分子成像。2013-2018年,美国威斯康星大学麦迪逊校区化学系,化学与生物工程系博士后。合作导师美国工程院院士Nicholas L. Abbott教授,美国科学院院士Samuel H. Gellman教授。研究方向:多肽的折叠结构与单分子力学研究。2018年至今,中国医学科学院基础医学研究所/北京协和医学院基础学院生物物理与结构生物学系工作。
