黄波教授早年在国外的研究方向为肿瘤免疫逃逸,主要研究对象为髓样前体抑制细胞(myeloid derived suppressor cells)。黄波教授发现在荷瘤小鼠骨髓中大量表达一类髓样细胞,并发现此类细胞可以明显抑制T细胞的抗肿瘤免疫应答,进而促进肿瘤的进展,相关工作发表在Cancer research杂志上,现已被引用1200多次,同时原创性发现肿瘤细胞通过表达TLRs,介导肿瘤免疫逃逸,相关工作也在Cancer research上发表,现已被引用450多次。 目前实验室汇集肿瘤免疫、肿瘤代谢、肿瘤生物机械力信号、肿瘤囊泡等肿瘤的核心方面,还原肿瘤、T细胞及代谢的真实面目,实验室的主要研究方向包括:1、肿瘤免疫:机理上阐明肿瘤免疫相关的根本性科学问题;采用多种手段和模型研究肿瘤免疫、肿瘤免疫逃逸的细胞和分子机制;肿瘤微环境中细胞、分子成分与抗肿瘤治疗相关机制;以及肿瘤转移前微环境形成的细胞和分子机制;2、肿瘤免疫治疗:以肿瘤细胞来源的微颗粒(囊泡)为载体,携带化疗药物、溶瘤病毒对肿瘤进行治疗;以及对微颗粒的化疗增敏效应或者作为新型口服疫苗进行研究;微颗粒治疗恶性肿瘤患者胸腹水的临床前机制研究;从肿瘤患者外周血中分离出肿瘤特异性T细胞并进行体外扩增3、医学生物力学:探究生物力学对细胞增殖、分化的影响,从而揭示生物力学信号在生物体内发挥作用的机制;运用生物力学筛选肿瘤干细胞,对肿瘤发生发展进行研究以及探寻肿瘤治疗新思路新靶点;阐明干性肿瘤细胞的物理力学特征;4、T细胞基本生物学:T细胞的记忆、胸腺的萎缩、干性T细胞的扩增等;5、代谢:回答糖原代谢的本质、为什么需要脂肪酸氧化、自由基的调控等核心问题。 实验室自成立以来在相关领域发表通信作者SCI文章90余篇,包括Science、Science Immunology、Science Translational Medicine、Cancer Cell、Molecular Cell、Nature Materials、Nature Immunology(4篇)、Nature Cell Biology(3篇)、Nature Biomedical Engineering(2篇)等国际主流期刊, SCI引用次数8000余次。
