"课题组围绕RNA代谢与造血调控,从RNA代谢角度解析干细胞命运转变、造血发育分化、疾病发生发展的基本规律,发掘RNA分子的新功能,鉴定RNA代谢的新机制,探索通过干预RNA代谢进行疾病治疗的新方法,并开展疾病的RNA治疗。近年来,取得多项重要发现,相关工作以通讯/第一作者在Cell Stem Cell、Developmental Cell、Blood、EMBO J(2015, 2020)、Nature Communications、Genome Research、Stem Cell Reports 等学术期刊发表,曾获教育部自然科学二等奖。获国家优秀青年基金(2021)、北京市“科技新星” (2017)、中国医学科学院“青年医学人才奖励项目”(2018)、“协和新星” (2015)等人才计划支持。 近年来,在相关领域取得多项重要发现: 1. RNA代谢与造血调控 利用单细胞测序技术绘制出造血干细胞发育全程的lncRNA图谱,揭示了调控“内皮-造血”转换的关键lncRNA(Cell Stem Cell 2019),文章发表后被Faculty 1000推荐阅读;同时首次描绘了成体造血干细胞分化层级的RNA编辑图谱,并从中鉴定了调控造血干细胞分化的关键编辑事件(Blood 2021);通过系统性调查假基因的表达特征及与疾病的关联,发现假基因HBBP1演化出调控人红细胞发育及疾病的重要作用及全新机制(Developmental Cell 2021)。 2. RNA代谢与干细胞命运 系统性揭示了RNA结合蛋白在胚胎干细胞多能性退出中的作用,发现hnRNPLL通过调控mRNA可变剪接促进胚胎干细胞退出多能性(EMBO J 2021);发现系列microRNA可协同转录因子调控造血干细胞及胚胎干细胞分化(EMBO J 2015);描绘了多能干细胞向造血所在中胚层分化过程RNA定位的动态图谱及规律(Stem Cell Reports 2024);揭示了假基因在人类早期胚胎发育过程中的潜在功能(J Genet Genomics 2024)。 3. RNA代谢与血液疾病 揭示了RNA代谢在血液疾病发生发展中的作用,鉴定了新的潜在治疗靶点;建立了正常血细胞及白血病细胞的RNA编辑数据库REDH(Chin Med J 2024);发现假基因HBBP1可通过提高TAL1 表达参与促进β-地中海贫血患者中胎儿血红蛋白水平升高,减轻了地贫患者的贫血症状(Developmental Cell 2021)。 "
